TLR3(又名CD283)是一个胞内受体,可识别与病毒感染相关的双链RNA(dsRNA)分子模式。其中一个TLR3的配体,Poly(I:C),是人工合成的dsRNA类似物。TLR3信号通路不依赖于MyD88,而是通过接头蛋白TRIF(包含TIR结构域的接头蛋白,可诱导IFN-β产生)发挥作用。IRF3(干扰素调节因子3)被磷酸化后导致IFN-β(β干扰素)产生,进一步活化ISRE(干扰素激活反应元件)。另外,TLR3信号通路的激活也可导致AP-1和NF-kB等转录因子入核,促进一些炎症因子的释放。
