炎症小体是一种多蛋白复合物，其活性与生理性和病理性炎症有关，可激活炎症性Caspases并激活IL-1家族的细胞因子（IL-1β、IL-18 和 IL-1α）。目前已经报道了几种含有不同传感蛋白的炎症小体，如NLRP1、NLRP3、IPAF、NLRP6、NLRP10、NLRP12、Pyrin、RIG-I 和AIM-2。这些炎症小体大多需要适配蛋白ASC（凋亡相关斑点样蛋白，含CARD）招募Caspase-1，Caspase-1被裂解和激活，导致其各种靶点被裂解，并引起炎症因子 IL-1β和IL-18的成熟和分泌。
NLRP3/NALP3属于NOD样受体家族，连接刺激信号与Caspase-1激活，是炎症小体的重要组成部分，在炎症调控中发挥着关键作用。越来越多的证据表明NLRP3/NALP3的突变会导致一系列自身炎症性疾病，如家族性寒冷型自身炎症性综合征（FCAS）、穆-韦综合征(MWS)和慢性幼儿神经皮肤关节综合征（CINCA）。
检测炎症小体激活的金标准方法是炎症小体Caspase-1的免疫印迹，但也存在一定的技术难度，特别是在蛋白质沉淀、蛋白质分离和转移方案的选择方面。另外，炎症启动和激活步骤也是免疫印迹实验成功的关键。
